Abstract
In order to investigate the effects of a calcium antagonist, verapamil on the renal function and renin release, experiments have been done in una- nesthetized rabbits. Agents were infused intrare- nally. Glomerular filtration rate, fractional excretion of urinary volume and free water clearance were increased. Urinary excretions of sodium, chloride, potassium, and fractional excretion of sodium were increased. The responses were dose depen- dent. Plasma renin activity was increased in the hypotensive dose of verapamil. Intrarenal arterial infusion of trifluoperazine resulted in increases of glomerular filtration rate, urinary volume, and urinary excretion of electro- lytes. The renal effects of verapamil were acce- ntuated under the influence of trifluoperazine- except glomerular filtration rate. The stimulus- response curves by the verapamil on the renal parameters showed left shifting with combining the trifluoperazine. The results suggest that the renal effects of verapamil infusion in unanesthetized rabbits may be caused by hemodynamic changes primarily, and the renal responses caused by the two agents may have different mechanisms of calcium depe- ndent processes beyond the intracellular level of calcium.
