본태성 고혈압 환자에서Perindopril 의 가압효과 및 안정성에 관한 임상적 고찰 |

Abstract |
In order to investigate the antihypertensive effect and the safety of oral perindopril, long acting angiotensin converting enzyme inhibitor, daily doses of 4 to 8 mg once a day were administered in 20 essential hyperten- sive patients whose diastolic blood pressures were between 95-115 mmHg. Blood pressure and pulse rate were measured every 2 weeks, The complete blood count with platelet count, blood chemistry by SMA-12 including serum electrolytes and urinalysis were perfor- med before the treatment and at the 6th week, and 12th week of therapy. Any kinds of side effects were actively questioned by physicians. The results are as follows: 1) The mean age was 49.2 years, and the male-female ratio was 9:11. 2) Blood pressure fell significantly in 2 weeks of treatment with peridopril (M±S.D., 156.0± 16.7 vs 131. 8±16.9, 102.8±4.7 vs 84.3±7.5, p<0,05), and during 12 weeks of the treatment with perindopril, statistically significant blood pressure changes were maintained. 3) There were no significant changes in pulse rate (72.4±4.8 vs 71.0±6.3, p>0.05) and body weight (63. 1±13.9 vs 66.1±4.6, p>0.05) during 12 weeks of treat- ment with peridopril. 4) There were no significant changes in blood chemis- try including blood urea nitrogen, serum creatinin, blood sugar, transaminase, uric acid, cholesterol, and electro- lyte. 5) There were no statistical differences in hematologic findings, but the amounts of urinary pro- tein were significantly decreased (p<0.05) at 12 weeks of treatment. 6) Total 5 patients (25 %) had various side effects: cough (15%), headache (10 %), constipation (5%), indiges- tion (5%), which were not so severe to discontinue perindopril. On conclusion, perindopril in dose of 4 to 8 mg once a day was effective in hypertension control and well toler- ated in most cases without severe side effects in essen- tial hypertensive patients with diastolic blood pressure between 95 and 115 mmHg. |
