Abstract
One of the mechanisms of the CyA-induced renal vasoconstriction is the altered prostanoids synthesis. Although there are many in vitro or experimental studies that demonstrated CyA-induced changes in prostanoids production, studies in renal transplant recipients, especially in the early post-transplant period when large doses of CyA are administrated, is rare. To investigate the changes of prostanoids production and renal hemodynarnics in CyA-treated renal recipients, 24 hour urinary excretion of PGF and TxB, and renal clearance of inulin(CgI) and PAH(Cpp) were deterrnined at 1 and 4 weeks post-transplantation in 9 renal recipients treated with CyA, who did not develop acute rejection or urologic complications during the study period. The ratio of urinary TxBz/PGFy, was significantly higher than norrnal controls(1.1±0.2, meanSE) at one(2.7±0.4, p<0.01) and four weeks(2.2±0.3, p< 0.01). Urinary PGF>. Tended to be lower at 1 week(39659 ng/day, p=0.067) and was significantly lower at 4 weeks(357±49, p<0.05) compared with normal controls(655 ±94). Urinary TxBz at 1 and 4W were not significantly different from that of the controls. Cr and CpAp> appeared to be low at both 1 and 4W. Between 1 and 4W, PGF, tended to decrease(p=0.083) but TxBy, TxBz/PGF>Cr and Cp, were not significantly changed. These results indicate that in renal transplant patients treated with CyA the ratio of TxBz/PGF, was increased, primarily due to the decrease in PGF
